Perform an Air Assassination on a Poisoned NPC - 20
The Poison Blade is given to Ezio in DNA Sequence 5 by Leonardo da Vinci, with Extra Poison being able to be purchased from Doctors around Italy.
The best way I found to do this is to distract a Guard with some courtesans, close to somewhere you can quickly climb, something like a Guard's Shelter. Then stabbing the guard with the Poison Blade, climbing up and air assassinating the Poisoned Guard.
